Suppose you were to carry out a mosaic analysis of the EGFR homolog let-23, using a homozygous let-23(-) strain that carries a small free duplication including the let-23(+) gene.  The duplication is occasionally lost during somatic development, producing a mosaic animal. The vulval precursor cells are derived from the AB lineage, while the entire gonad is derived from the P1 lineage. What phenotype, Vul or normal, would you predict if the duplication were lost in P1 but retained in AB?  If it were lost in AB but retained in P1?  How is this gene product behaving—in a cell autonomous or non-autonomous fashion?

Making the vulva requires that a lin-3 encoded signal from the gonadal anchor cell is sensed by a let-23- encoded receptor on the VPC P6p.  The gonad is derived from P1; the VPCs from AB. Therefore, vulva formation will work fine without let-23 expression in the AC (ie, normal when let-23 is lost in P1), but will require let-23 in the VPCs (thus Vul when le23 lost in AB lineage that makes the VPCs).  It’s autonomous—it’s a receptor.
