Both AFLP and rep-PCR are DNA profiling methods that assay for variations in DNA sequence within a genome, rather than ribotyping, which focuses exclusively on only on the differences of one particular gene. The rep-PCR method uses highly conserved repetitive sequences to amplify. AFLP involves one or two restriction enzymes to digest genomic DNA and selectively amplifying digested sequences. Both require the DNA to be resolved with gel electrophoresis, and the band patterns serve as "fingerprints" for comparative purposes.
