Problem 12.27
A wild-type culture of haploid yeast is exposed to ethyl methanesulfonate (EMS). Yeast cells are plated on a complete medium, and 6 colonies (colonies numbered 1 to 6) are transferred to a new complete medium plate for further study. Four replica plates are made from the complete medium plate to plates containing minimal medium or minimal medium plus one amino acid (replica plates numbered 1 to 4) with the following results:

Part A
Identify the colonies that are prototrophic (wild type).

Wild-type organisms can produce everything in the biological pathway that they need to survive, therefore, they can grow on medium that is not supplemented with any other nutrients or amino acids.

Mutants are lacking portions of the biological pathway that allow them to grow. Therefore, they rely on external supplements to grow. For instance, if you have a vitamin B deficiency and you begin taking vitamin B, your body starts functioning better. If you have a vitamin B deficiency and begin taking a vitamin C supplement, your problem won't go away.

How can you relate the above example to this problem?
I hope this helps!
