There are two proteins; one has a molecular weight of 10,000 Dalton and a pI of 7.1. The other has MW of 40,000 Da and pI of 4.9. They are separated by two-dimensional gel electrophoresis. The first dimension is PAGE. The second dimension is on a pH gradient gel for isoelectric focusing. Sketch their likely relative position at the end of the gel electrophoresis.

Could you tell me the reason why the one that has heavier molecular weight
is on the above? Shouldn't it be below because it's heavier?

The heavier the molecule, the shorter the distance it will travel. Think of something heavy running really quickly - won't go far. Whereas the smaller molecule will run farther and quicker.

Yeah! I got your point that the smaller molecule would travel further.
But, what I saying is that the position of both molecules, which is above and which is below.
