Initial rate increases linearly as the substrate concentration increases as the active of site the enzyme becomes more likely to collide with the substrate molecule forming an E-S complex. As the substrate increases a point will be reached where adding more substrate has no effect on enzyme activity - this is when excess substrate is present and the amount of enzyme is not limiting.
