I have done some research on prion proteins, and I have heard that they "die" after a year under most circumstances. How exactly does this happen? Could they be reactivate many years(possibly decades) later or would the protein components be long gone for anything to happen?

They still have not found out why they actually die or why they collect in nerve cells to die. Once a protein unfolds it cannot refold. Their is still a lot of mystery surrounding prions that they are looking for answers. They are not even sure how they cause the cascading effect within the body creating more prions which lead to several diseases. The one's that are dead are dead. Your lysosomes recycle the dead matter back into the cell.
