Receptor serine/threonine kinases act through receptor-regulated Smads and the accessory Smad4. Together, they enter the nucleus as a complex following receptor activation.

Ras is regulated by GAPs and Sos (guanine-nucleotide exchange factor). This in turn activates a cascade of phosphorylation events, resulting in the activation of transcription factors that mediate gene expression.
