Since bacteria do not process RNA or proteins the way we do, a split gene is more difficult to clone and express. The split gene can be cloned normally, but its expressed product will most likely be wrong. To compensate for this, the inserted DNA should be cDNA, which will not have the introns, since it was created from mRNA.
