If we want to grow a bacterial clone with a plasmid that carries a desired DNA fragment inserted within the antibiotic ampicillin resistance gene, we should not grow it in a medium containing ampicillin.

That is false.  A medium containing ampicillin would select for clones that contain the plasmid with the ampicillin resistance gene.

your question isn't too clear, but to get the correct clone of the PLASMID not the bacterium. you must cut out the desired DNA fragment using a specific restriction enzyme, then the plasmids should be put into DNA lygase and agar to allow the DNA fragment to join back into plasmids and more to grow via mitosis, now the agar plate shoudl be put into ampicillin to determine which of the cultures of plasmids have managed to fully recieve the ampicillin resistance gene
