Title: Formamide in hybridization buffer is w/v? or v/v?

Greetings, good friday.

I have a question about hybridization buffer..

actually, you know, hybridization include salt(SSC), detergent(SDS), and formamide. but SSC is selling by stock sol(20X) so we inject when calculate diluted vol, SDS is solid powder then % is (w/v). but formamide is liquid, some sells unit by ml, others sells unit by g... So I confused it is (w/v) or (v/v).

when you add formamide in hybridization buffer, you use (w/v)%? or (v/v)%?

That depends:

When the solute is a liquid sometimes it is convenient to express its concentration in volume/volume percent (v/v %). The definition of v/v % is

Weight/Volume (% w/v) is the weight in grams of a solute per 100 milliliter of a solution. For example, a 50% w/v solution of PEG 6000 is made by adding exactly 50 grams of PEG 6000 solid to a total, final volume of 100 milliliters. NOT by adding 50 grams of PEG 6000 to 100 milliliters of water.
