Title: Help i have absorbance of LDH enzyme vs time. how do i change the absorbance per minute?
Post by: roaringspring on Jun 11, 2013
i have to find enzyme activity but i have to change absorbance per min  first. i have absorbance  every 10 seconds. what i have done is divide the 10 sec by 60 and then divide 20 by 60 and so on.is it alright.

peter s is corrrect (and his avatar and name look alarmingly like mine!  weird!)

Id add that you should look up Lineweaver/Burke Plot on wiki or something...
see peter S is correct, however I think I remember enzyme activity being logarithmic... it's first minute or 2 are VASTLY faster than the next several minutes after... this is because the enzyme is becoming "limited" and needs more substrate to continue at its current speed.  I vaguely remember the L-B plot being a way to calculate it Abs/Min... been a while since I did the LDH lab in undergrad biochem tho.
