The following shows a restriction endonucleases cleave DNA. abbrev R represents the purines (A&G) and pyrimidines (C, T & U) are abbrev Y. The abbrev W represents A or T.

Enzyme: PpuMI
Targe Sequence: 5' RGGWCCY 3'
                            3' YCCWGGR 5'

Cleavage: 5'RG           GWCCY 3'
                 3' YCCWG          GR 5'

Question: Plasmid DNA and a gene of interest are cut with the enzyme PpuMI. Write a possible sequence of bases for the sticky end of the gene in the 5' to 3' direction.

Hi alatq33, thos elook like palindromic sequences right?

Just out of curiosity, what textbook are you using?

5' - RG
        GWCCY - 3'

       GWCCY - 3'
5'- RG

Pseudomonas putida is the genomic source for PpuMI
