Title: Why dilute ligation mixture before transformation?
Post by: adriata on Mar 15, 2017
I understand that diluting the ligation reaction mixture prior to transformation increases the transformation efficiency, but why is it that less DNA yields better results in this case?

At high [DNA] you also could have an increased amount of 'impurities' (salts, etc) that can affect transformation efficiencies.
