Prior to vaccination, the patient's skin was sanitized with alcohol.

Alcohol cannot sterilize.

Desiccation and cold only temporarily inhibit the growth of microbes and these micorbes can become active again when proper growth conditions are restored. Some microbes can in fact be preserved by such methods, thus extending the likelihood of their reactivation over many months.
