Activity 4: Determining the Effect of Propylthiouracil
on Metabolic Rate (p. 48)
7. On the normal rat, the metabolic rate after propylthiouracil injection is slower than the
baseline metabolic rate.
Propylthiouracil is antagonistic to thyroxine and will tend to decrease the effects of
thyroxine.
On the thyroidectomized rat, the metabolic rate after propylthiouracil injection is the
same as the baseline metabolic rate.
Since the thyroidectomized rat cannot make any thyroxine, the propylthiouracil has
nothing to be antagonistic to and therefore has no effect.
On the hypophysectomized rat, the metabolic rate after propylthiouracil injection is the
same as the baseline metabolic rate.
Since the hypophysectomized rat does not have a functional thyroid gland, no thyroxine
is being made and there is nothing for the propylthiouracil to be antagonistic to.

This is probably a bit late but Activity 1 #12 is not given in duddy's answer key because you have to use data that you have collected and recorded in chart 1. After you convert the weight data from grams to kilograms use the formula given to determine the metabolic rate. These answers will vary depending on what you have on your chart.

Exercise 4: Activity 4:  Determining the Effect of Propylthiouracil on Metabolic Rate

1.   What was the effect of propylthiouracil on the normal rat’s metabolic rate?  It caused the rate to decrease. How does it compare to the normal rat’s baseline metabolic rate?  The metabolic rate of the baseline rat was significantly higher
2.   Why was this effect seen? Injected with tyroxine caused an increase in grams and ml of O2 levels, because the tyroxine is a hormone from tyroid gland which increase the basal metabolic rate.
3.   What was the effect of propylthiouracil on the thyroidectomized rat’s metabolic rate?  There was no effect. How does it compare to the thyroidectomized rat’s baseline metabolic rate?  The metabolic rate was the same for the two (2) rats.
4.   Why was this effect seen?  The rat needs both the pituitary glands as well as the thyroid gland to produce thyroxine. PTU inhibits the production of the thyroxine, but thyroxine is already not being produced in this rat due to the absence of the pituitary and/or thyroid glands.
5.   What was the effect of propylthiouracil on the hypophysectomized rat’s metabolic rate?  There was no effect. How does it compare to the hypophysectomized rat’s baseline metabolic rate?   The metabolic rate of the baseline rat was the same for these two (2) rats.
6.   Why was this effect seen?  Because propylthiouracil is an anti-thyroid drug used to treat hyperthyroidism by decreasing the amount of hormone produced by the thyroid gland and the rat had the thyroid gland removed.
