Denaturation of a protein occurs when you disrupt the secondary, tertiary and quaternary structures of a protein. This can occur by the use of heat, detergents, pH changes, alcohol, etc.

Heat and alcohol can be used to disrupt hydrogen bonds and non-polar hydrophobic interactions. Hydrogen bonds are responsible for holding together the secondary structure of a polypeptide. pH can affect the chemistry of the amino acids and their residues in the tertiary structure, breaking "salt-bridges". The ionizable groups in amino acids are able to become ionized when changes in pH occur. A pH change to more acidic or more basic conditions can induce unfolding.
