To determine whether a specific DNA fragment in yeast is present in wild type versus mutant DNA .. which technique should I use?
a)digest genomic dna with restriction enzymes and run on gel and compare band sizes
b)perform southern blot
c)perform western blot
d) perform northern blot
e)perform reverse transcription-PCR
f)a and b
g) a and d

a and b (technique a is necessary for technique b).

c) is a procedure specifically for proteins so is not applicable.

d) is a procedure specifically for RNA so is not applicable.

e) might be possible if the mutant piece of DNA is long enough for a PCR. If it was small e.g. 40 bases then PCR would be impossible.
