What purpose does the kaleidoscope marker serve in your coomassie-stained gel? If you were doing a western blot, how would the kaleidoscope maker be useful?

kaleidoscope marker is used to visualize the migration of proteins.It provides reference points on the polyacrylamide gel. It marks the specific distance, where protein of specific size is expected.

The coomassie-stained gel belongs to non-denaturing electrophoresis. Proteins remains in a folded state and they run through the gel with different rates , in the presence of charge. The rate depends on the various factor like composition of amino acid in the protein and mobility of the protein size/shape in the gel . The kaleidoscope marker serve as a indicator whether protein transfer is complete after the 30 minute transfer period on the nitrocellulose filter or not.

This marker is used in the western blot to estimate the molecular weight of the proteins on the gel.
