GPI-anchored proteins are synthesized:

1. as transmembrane proteins, cleaved, and attached to the GPI group on the lumenal surface of the ER.
2. on free ribosomes and attached to the GPI group on the inside of the ER.
3. completely into the lumen of the ER and attached to the GPI group on the inside surface of the ER.
4. on free ribosomes and attached to the GPI group on the outside of the ER.
5. as transmembrane proteins, cleaved, and attached to the GPI group on the cytosolic surface of the ER.

GPI-anchored proteins are synthesized as precursors with a cleavable, hydrophobic amino-terminal signal sequence that targets the protein to the lumen of the endoplasmic reticulum (ER) and a cleavable, carboxy-terminal signal sequence that directs GPI anchoring.
