Microarray analysis using DNA Chips and Southern blot analysis both rely on a fundamental property of DNA that is the basis for their utility.  What is that property and why does this property cause temperature to be a critical factor in the performance of these types of analyses?

I think the property they are referring to is the fact that the two strands of DNA are held together by hydrogen bonds. The more hydrogen bonds there are, the higher the amount of heat is required to separate the complimentary strands.
