Cloning and transformation experimentYou are about to start an experiment which requires you to clone PCR products of your favorite gene (YFG) into a plasmid and transform it into E. coli. In checking your lab freezer, you find that the following resources are available to you:
The antibiotics kanamycin and ampicillin.
Plasmid vector pA, which includes a kanamycin resistance gene.
Plasmid vector pB, which includes a tetracycline resistance gene.
E. coli cells of strain X, which is resistant to kanamycin but sensitive to both ampicillin and tetracycline.
E. coli cells of strain Z, which is resistant to ampicillin but sensitive to both kanamycin and tetracycline.
In order to select ONLY for transformed E. coli colonies containing the plasmid,which one of the following combinations of plasmid vector, E. coli strain, and antibiotic could you use. a. Clone YFG into plasmid vector pB, transform your plasmid into E. coli strain X, and plate onto media containing ampicillin.
b. Clone YFG into plasmid vector pB, transform your plasmid into E. coli strain X, and plate onto media containing kanamycin.
c. Clone YFG into plasmid vector pA, transform your plasmid into E. coli strain X, and plate onto media containing ampicillin.
