One of the 20 naturally-occurring amino acids. Structurally and chemically unique as the only one of these to have a secondary amine functionality; actually it is a cyclic amino acid. The amide of proline in a protein backbone thus has no hydrogen to serve as a donor in hydrogen bond formation. Proline is particularly abundant in collagens, a group of connective tissue proteins. A significant number of proline residues in collagens are post-translationally modified to 4-hydroxyproline (Hyp). The hydroxylation is carried out by prolyl hydroxylase, an iron-containing dioxygenase that utilizes ascorbate (more commonly known as vitamin C) as a reducing cofactor.
