Description
The lengths of unwound DNA and DNA–RNA hybrid were originally estimated from reactivities of transcription complexes with reagents such as KMnO4, which oxidizes bases in single-strand nucleic acids.

The length of DNA in contact with the enzyme is determined by footprinting.

Six or seven nucleotides of RNA behind the DNA hybrid are protected from ribonuclease attack by binding to the enzyme.
