The vaccinia virus itself does not cause disease in humans and is therefore an excellent virus to administer alive, containing recombinant proteins for immunization against a particular disease. A plasmid containing a partial sequence of vaccinia virus housed within E. coli is used to homologously recombine with the vaccinia virus. In this way, genes from pathogens can be inserted into the plasmid in E. coli, and the virus will express the proteins to immunize an individual from the particular pathogen.
