The western blot test is the best test to diagnose infection with HIV. It is performed in three steps. First, electrophoresis of the antigens in a solution is carried out. Next, the protein bands from electrophoresis are blotted and transferred to a nitrocellulose membrane that is cut into strips. In the final step, ELISA is performed on each nitrocellulose strip by incubating it with the serum sample from the patient being tested.
