So your are conducting a research project and need to insert a gene from one organism into a plasmid.  There are numerous restriction enzymes available to use and you need to choose an appropriate one.  Explain two things you should consider when making your decision.

You want to look for two unique restriction sites so that there is one place and only one place where your gene will be inserted into the plasmid.

Secondly, you want to make sure that neither of these restriction sites are in other important regions of the plasmid. For example, if you plopped your gene into the middle of the origin of replication, your plasmid would not function. Also, your gene does need to be immediately after whatever inducible promoter region you are using (generally the lac promoter).
