Hi all when I was working in lab I got doubt. I had to do soil serial dilution and perform spread plate technique. I need to know that, for soil serial dilution how much gm of soil should be taken  and mixed with how much ml of distilled water and the same question is for Liquid sample serial dilution. Kindly provide me some website which has the needed details. Thank you.

For serial dilutions, you usually do a series of 1/10 dilutions. For instance, you would homogenize 1 g of soil in 9 mL of diluent (distilled water), then take 1 mL of the blended soil suspension and add to 9 mL of distilled water. Typically, you'd do a series of these to make sure that one of the plates has a countable number of colonies (between 30-300 colonies); typically, a series of at least 5-6 dilutions is plated. For liquids, you'd do the same thing, but add 1 mL of your sample to 9 mL of diluent.
