For our new project we're aiming to date blood stains found within a forensic crime scene. Several publications (listed below) have reported working methods for dating those blood stains. These mainly rely on spectroscopy and the hemoglobin (and other forms) compound. We would like to be initiative and setup some new kind of procedure. We thought about a few things (if you have other ideas, please please let us know!):
  • Try to seperate the HbO2 molecule from the met-HB molecule (the difference is just Fe2+/Fe3+) using ion chromatography/protein seperation? (or some kind of chromatography) and detect them. (Maybe an other option would be a DAD, to detect at different wavelenghts.. however this is kind of the same as previously done in literature)
  • Bind different antibodies to the different forms of hemoglobin and calculate the ratios of these

We are kind of stuck and would like ideas, feedback, everything you can think of. Also other ways apart from hemoglobin to age blood stains are really really appreciated. Thank you very much in advance.

Publications
Hyperspectral imaging for the age estimation of blood stains at the crime scene (2012)
Identification and age estimation of blood stains on colored backgrounds by near infrared spectroscopy (2012)
Age estimation of blood stains by hemoglobin derivative determination using reflectance spectroscopy (2011)

Really appreciate the help Padre, the only problem is that we cannot use any biological/molecular techniques so both literature references are worthless for me  Frowning Face
