Suppose you used 3 microliters of pGLO (0.03 microgram per microliter), and spread 250 microliters of a total reaction volume of 500 microliters.  If you count 100 transformed colonies, what is your transformation efficiency (TE)?

From what I remember what to do is Transformation Efficiency= # colonies/ ug pGlo per plate

A) 2.2 x 10 ^ - 3
B) 222
C) 2.2
D) 2.2 x 10 ^ 3

But the answer I am getting isn't close to these but 1.6, the paper I have to help me with this was the pre-lab that I turned in.

I get D). You had 0.09 mcg of DNA in the 500 microliter mix. You plated half of that and saw 100 colonies, so you must have had 200 transformants in the mix. 200/0.09 = 2222, which is about 2.2E3.
