I'm trying to culture diatoms and I am unsure how to mix the agar. I have liquid ag diatom growth medium, powder agar, gentamicin, and metacilicate. I'm wondering if I should use distilled water and boil it with the powder agar before mixing everything together--or do I use the liquid media and boil it directly with the agar powder to create the agar plates and then add the gentamicin and metacilicate?

It's kind of like making JELLO. You boil water and mix the ajar powder into it with the growth medium.
