Diphenhydramine is an antihistamine. This drug acts as a full agonist at the HA receptor, as part of an inflammatory reaction that bind to histamine. The histamine is released by the immune cells, which are mast cells and basophil cells, after the binding with IgE antibodies present on these cells with the allergens. Diphenhydramine works by inhibiting histamine from binding to this receptor, reducing the negative symptoms associated with histamine HA-receptor binding.

Methylprednisolone is a corticosteroid medication used to suppress the immune system and decrease inflammation. Unbound glucocorticoids cross cell membranes and bind with high affinity to specific cytoplasmic receptors, modifying transcription and protein synthesis. By this mechanism, glucocorticoids can inhibit leukocyte infiltration at the site of inflammation, interfere with mediators of inflammatory response, and suppress humoral immune responses.
