First, you obtain a thin section of the plant tissue containing the vascular bundle of interest. Use a dissecting microscope that explodes the image to 10x. If you have calipers, squeeze the tissue between the jaws of the caliper. This will tell you the diameter of the specimen. If you don't have calipers, a plane ruler can also do the trick. Beyond this, I'm not sure what sort of procedure you're looking for. Please let me know if you need further details, apologies for misunderstanding
