Studies of adopted children and twins support the notion that major depression is a moderately heritable disorder.
It is unlikely that a single gene is involved.
It is likely that a genetic predisposition interacts with stressful events to create the disorder.
A gene called 5-HTT comes in two forms, a long and a short form.  The short form appears to be related to depression.
Genes may exert their effects by affecting levels of serotonin and other neurotransmitters or by affecting production of cortisol.
