I know pepsin works best in acidic environments of around pH 2-3, but is there any other reason that makes it unsuitable to add?

The question makes it sound as if the pepsin will adversely affect the detergent mixture...or is the optimum pH the only reason?

Pepsin needs an acid environment to work.  The optimum pH is around 2.
