(a) This is usually achieved by precipitating chromosomal DNA from an alkaline lysate by neutralizing with potassium acetate.  Plasmid DNA remains soluble.

(b) Cells can be made competent by several different chemical treatments and incubated with plasmid DNA, or plasmid DNA can be introduced by electroporation, which gives the highest efficiency of transformation.
