in this y1 is 3, x1 is 4, y2 is 0, and x2 is -6, so the slope is -10 over 3 or -10/3

It doesn't matter, as long as you are consistent. Conventionally, you would treat the (x,y) pair with the smaller x value as (x_1,y_1) and the other as (x_2,y_2). However, it doesn't matter, because the ratio (y_2 - y_1) / (x_2 - x_1) is the same as the ratio (y_1 - y_2) / (x_1 - x_2), it's just that the sign of the top line changes but so does the sign of the bottom line.
