Why is it necessary to rinse the smear excessively with water?  Certain people said that, just need some amount of water just to remove its stain.  Is that true?

Gram-positive bacteria in purple colour while gram-negative bacteria is in red colour.  Is it because of the peptidoglycan present in the cell wall?

Can the thickness of peptidoglycan effect the outcome of this experiment?
