Curare blocks the receptor for acetylcholine, which acts as a neurotransmitter in certain cells to bring about an action potential in them. For example, skeletal muscle cells reach threshold after acetylcholine binds to the receptor site, which causes the sodium channel to open and depolarizes the cell. Curare blocks the receptor so acetylcholine cannot bind, which means the skeletal muscle cell cannot open the sodium channel and depolarize to reach threshold and make an action potential.

That's just one example but many other neurons and cells use acetylcholine to depolarize.
