Cell culture would be useful, but CANNOT replace human testing since effects can depend on the interaction between cells of different types.

Eventually you have to give the drug to humans, if only because you start selling it to treat illness.

Cell culture testing can reduce the risk of unforseen side effects the first time you give it to humans, but it cannot eliminate it. You would need to have cultures of every type of cells in the body, then cultures of each possible pair of types, then each possible triad, etc etc etc.
