Not knowing what the original mix was, not sure what you're asking. But if we start with the 2.2 ml (assuming nothing happened before that), then the dilution factor will be 3 because you took 1 ml, added it to 2 ml for a total volume of 3 ml.  That's a 1:3 or 3 fold dilution of the original 2.2 ml reaction mix.
