A DNA sequence encoding a five amino acid polypeptide is given below. The correct protein coding sequence contains a start codon, four amino acid codons, and a stop codon.

Locate the sequence encoding the five amino acids of the polypeptide, and identify the template and coding strands of DNA.

a)   The top strand is the coding strand. The bottom strand is the template strand.

Is this from Griffith's 10 ed Genetics?  I seem to remember doing this question when reviewing for the final for UC Berkeley.

The amino acids can be encoded by the codons given below:

Met = ATG
His = CAT or CAC
Arg = AGA or AGG or CGC or CGT or CGA or CGG
Lys = AAA or AAG
Val = GTT or GTA or GTC or GTT
Gly = GGA or GGC or GGT or GGG

Hence, the number of sequences possible (with possible combinations to give the desired peptide chain) are:

Met-His-Arg-Arg-Lys-Val-His-Gly-Gly
1*2*6*6*2*4*2*4*4 = 18432
