In E. coli, how are the Okazaki fragments of the lagging strands joined to create a complete DNA duplex?

Ans: Each Okazaki fragment has an RNA sequence at the 5 end and a free 3-OH end adjacent to the 5 end of the next fragment. In E. coli, DNA polymerase I replaces the RNA primer with DNA. The nicks between fragments are resolved by the enzyme DNA ligase, which joins the 3-OH end of one fragment to the 5-phosphate end of the next fragment.
