Help with FACS

I was wondering if it would be possible to look at cellular changes in mRNA expression in a specific cell type using FACS after acute slice pharmacology treatment? I haven't seen any papers that attempt using acute slice pharmacology before cell isolation, only drug treatments in the live animal. Is this sort of treatment implausible? If so, why? Just thinking out lout and would like some feedback.
