The only way to get neurotransmitters or synthesis enzymes to the presynaptic terminal of axons would be diffusion. This would be a slow process, so the cytoskeleton is needed to move the neurotransmitters and enzymes. No movement would limit the length of axons, which may be a meter or so long. No axon transport system would mean short axons and many more neurons with their axons needed to complete a neural pathway.
