No, amylase doesn’t use cellulose as a substrate. Starch is the substrate for amylase. Test Tube #3 should not show a positive Benedict’s test because cellulose is not the substrate for amylase.

The bacteria were able to digest the cellulose because they produce the enzyme, cellulase.
