How did von Ehrenstein et al. demonstrate that the specificity of codon recognition lies in the tRNA molecule and not in the amino acid it carries?

They chemically converted the cysteine on tRNA.Cys molecules to alanine in vitro. This Ala-tRNA.Cys was used to synthesize hemoglobin, which normally contains a cysteine on each ? and ß chain. The resulting hemoglobin molecules contained alanine at the position normally occupied by cysteine, which indicated that the tRNA had read the cysteine codon and inserted the alanine it carried.
