What is up promoter mutation ? How or why does it increase the number of mRNA made during transcription? What is a down-promoter mutation and how does that reduce mRNA transcription?

An up mutation is just the general term for a mutation that causes an "upping" in transcription rate.  Usually it makes the promoter more easily bindable to the transcription factors need to begin transcription.  A down mutation has the opposite effect and lowers the affinity of the transciption factors for the promoter region, lowering transciption rates.
