I am master student I have E.coli Xli.Blue strain and I need to improve a protein folding, and good growth of the bacteria.  I use in my experiment 1.25ml of 0.5MDTT and i got no growth at all which means high dose I used or problem with the DTT solution as it is unstable. So please help me to find a good range of DTT concentrations to try.
Any answer will be appreciated.

Typical working concentrations of DTT in buffers for protein work range from 1 - 10 mM.  But the optimal concentration for any particular protein is protein specific so your best bet is to set up several different concentrations and see which one works best.  If you see a range of activity you can repeat with various doses closer to the one that worked better and repeat until you get the concentration you want.
