I'm doing a lab that has to do with agar diffusing in NaOH. My independent variable is the temperature of agar, chilled and room temperature. My dependent variable is the percent diffusion.

Any help would be appreciated.

To start off, I think you mean diffusion of say a plasmid into bacteria, because an agar is the plate bacteria get put on. Diffusion can be done by, in your case, through heat shock. Heat shock allows for foreign DNA to enter bacterium by permeating the membrane, so I suppose you could say that with increase in temperature your percent diffusion rises. However, too much heat denatures alot of proteins so that tampers with data.
