We tested a new restriction enzyme on a plasmid of 6.3kb. The digest was run in an agarose gel against linear standards. But only a single band was present in the plasmid digest lane and when its size was calculated from the standard curve, it was found to be only about 2kb. (excluding experimental error) i am not usre why?

I'm assuming that the restriction enzyme must have "cut" the plasmid in 3 different areas evenly spread throughout the genome (i.e. 6.3 divided by 3 = 2.1 kb).
